Abstract

Pain was the most common problems found in the body. And pain has considered as a sign of the beginning of a problem in the body such as inflammation. Inflammation was a normal response to injury caused by a tissue of physical trauma, chemical substances that damage, or microbiologic substances [1]. Salicylic acid derivates were one of the analgetics that had been used to treated pain, but there was side effects such as gastric irritation. Therefore, 4-chloromethylbenzoyl salicylic acid had been synthesized from 4-chloromethylbenzoyl chloride and salicylic acid by acylation reaction and based on Schotten-Baumann method [2]. The yield of 4-chloromethylbenzoyl salicylic acid was 83.1%. In this research, analgesic activity was tested using writhing test method. The analgesic activity determined by perceiving degradation amount of writhe frequency moreover calculated pain inhibition percentage and ED50 value. The result showed that 4-chloromethylbenzoyl salicylic acid had EDso 11.3 mg/kg BW, while acetylsalicylic acid had ED50 20.8 mg/kg BW. That result was caused by the addition of 4-chloromethylbenzoyl in the structure, it caused the increase of lipophilicity and penetration on biologic membrane, so the amount of drug that interacted with receptor were increased. In conclusion, 4-chloromethylbenzoyl salicylic acid had higher analgesic activity than acetylsalicylic acid.
