Abstract

Latar Belakang: Diabetes melitus masih menjadi masalah kesehatan dunia. Salah satu komplikasinya adalah infertilitas pada pria akibat hiperglikemia dan kondisi stres oksidatif. Umbi porang. Tujuan: Mengetahui efek ekstrak umbi porang terhadap mikrostruktur testis tikus DM. Metode: Penelitian ini menggunakan metode True Experimental dengan rancangan Post-Test Only Control Group Design. Hewan coba yang digunakan untuk penelitian ini adalah Rattus norvegicus strain Wistar jantan yang diinduksi aloksan. Jumlah sampel yang digunakan adalah 30 ekor Rattus norvegicus yang dibagi menjadi 6 kelompok, yaitu kelompok N, K+, K-, P1, P2, dan P3. Rattus norvegicus diinduksi aloksan 125 mg/kgBB secara intraperitoneal. Kelompok K- diberi kelompok K+ diberi sedangkan kelompok P1, P2, dan P3 diberi ekstrak umbi porang dengan dosis 100 mg/kgBB, 200 mg/kgBB dan 400 mg/kgBB. Ekstrak umbi porang diberikan pada Rattus norvegicus dengan cara sonde selama 50 hari. Hasil: Analisis uji statistik dengan menggunakan metode uji ANOVA dan uji Kruskal-Wallis. Rerata skor tahap spermatogenik pada kelompok K- 6.82, kelompok P1 8.06, kelompok P2 8.20, dan kelompok P3 8.96 dengan hasil signifikansi uji hipotesis (p = 0.000). Rerata jumlah sel Leydig persepuluh lapang pandang pada kelompok K- 20.06, kelompok P1 21.90, P2 25.92, dan P3 32.82 dengan hasil signifikansi uji hipotesis (p = 0.000). Rerata ketebalan epitel tubulus seminiferus pada kelompok K- 49.81, kelompok P1 52.75, kelompok P2 49.69, dan kelompok P3 53.56 dengan hasil signifikansi uji hipotesis (p = 0.603). Simpulan: Jumlah sel spermatogenik dan sel Leydig sesudah pemberian ekstrak umbi porang mengalami peningkatan namun ketebalan epitel tubulus seminiferus tidak didapatkan perbedaan yang bermakna. Dosis ekstrak umbi porang yang optimal untuk meningkatkan jumlah sel spermatogenik dan sel Leydig adalah 400 mg/kgBB.
