Abstract

Orally Disntegrating Tablet (ODT) is tablets that dissolve in mouth with help of saliva so easily swallowed without using water. ODT being produce using direct compression metode. The choosen of this method cause of need a short time, but no one of the material showed an ideal characteristics. Therefore, it maked by co- process metode. This study aims to determine the effect of a mixture of concentration PVP K-30, crospovidone, and mannitol on the characteristics of the excipient co-process, to determine the concentration of a mixture of PVP K-30, crospovidone, and mannitol excipient to produce optimum co-process with methods factorial design, and to determine physical properties of domperidone ODT tablets are compressed with excipients co-process optimum. In this study, factorial design was used with three factors and two levels as the method for determine formula. The factor was PVP K-30 combination low-level used 5% concentration and high-level concenteration used 10%, for crospovidone, low-level used 2% concentration and high-level used 8%, and for mannitol, low-level used 5% concenteration and high-level used 10%. Responses determine which formula being used to co-process optimum were Carr’s index, Hausner ratio, hardness, friability, disintegration time, wetting time, and ratio of water absorption. The results of research showed that PVP K-30 concentration, crospovidone concentration, mannitol concentration, and the interaction has a different effect of each respon. Using design expert program, the optimum formula of co-process material is a combination of PVP K-30 (10 %), crospovidone (8%), and mannitol (10%). Wetting time was the only respone that showed significant difference. The physical properties of domperidone ODT tablets are compressed with excipients co-process optimum qualifies as ODT tablets.
